Features Under physiological conditions ubiquitylation does not target BRCA2 to proteasomal degradation, as shown by treatment with MG132 proteasomal inhibitor. However, treatment with Mitomycin C (MMC), the DNA interstrand cross-linking agent, increases ubiquitylation level and leads to proteasomal degradation of ubiquitylated BRCA2.

Comments BRCA2 specifically forms complexes with a deubiquitylating enzyme USP11. USP11 can deubiquitylate BRCA2 in vitro or in overexpression assay, but not in vivo. BRCA2 may play a role in recruiting USP11 to its physiologic substrate.
